In addition to JB4 and fuel pump upgrade (stock turbo), what are the minimum upgrades needed to run E85 from the pump?

Also need the flexfuel wires for the JB4. I'm assuming you already have all the bolt on mods like intercooler, intake, exhaust, etc.? E85 is just another type of fuel. To make any more HP, you'll need to introduce the extra airflow to match the potential of the octane benefits of E85.

You can run an e30 mix on stock lpfp. you can run e50-e60 on upgraded lpfp, you can run full e85 with upgraded lpfp and hpfp or upgraded lpfp and port injection!
